I added the code to my theme’s functions.php file but it does not working, %excerpt% is shown, If I edit the post’s meta manually that post’s %excerpt% is not shown in meta. Otherwise all post shows the %excerpt% meta not the Single Post Description that I set from Rank math plugin Title & Meta option.

Could you please share the affected URL and the excerpt so we can check? Also, please share the default description in Rank Math > Titles & Meta settings.
Meanwhile, please note that the filter may not be visible while editing the page as it only works when it loads in a browser. You can check the published/saved meta description via its source code.

When I inspect the code it show the description, but when I edit the post then under the Rankmath option, there showed the excerpt ৳32,000 only

The excerpt of the post takes priority over the global description. Since you have applied the filter, the correct description will be added to the front end but the preview in the meta box will show the excerpt. It doesn’t affect the SEO of your site as the correct description is there in the front end and the search engines will see that.
